Here we describe the isolation of the gene of a variant of dermcidin, designated as DCD-α. This variant has 75% homology to dermcidin and has been found in an isolated population in the Central Pacific Basin only. This DCD variant possesses 15 extra amino acid residues that are not found in the native form. Specifically, it consists of 125 amino acids in its precursor peptide and 63 amino acids in the processed form. The proteolytic cleavages of the precursor by kininase followed by kallikrein give rise to the mature peptide, DCDα1. DCD-α1 has absorbance at the 400nm and 700nm regions of the visible light spectrum and is viewed as green. The colour of the peptide is most intesnse at pH 5.5. We speculate that DCD-α1 may possess similar anti-microbial properties as its native form. However, futher experiments are needed to elucidate the antimicrobial activities of DCD-α1 Jedy Go§, Madonna A. de Lemos,§ Joanna Ip,§ Wei-Hung W. The multiple facets of dermcidin in cell survival and host defense.
Eccrine sweat glands, which are distributed over the whole bodies of primates and humans, have long been regarded mainly to have a function in thermoregulation. However, the discovery of dermcidin-derived antimicrobial peptides in eccrine sweat demonstrated that sweat actively participates in the constitutive innate immune defense of human skin against infection. In the meantime, a number of st...
متن کاملNaturally processed dermcidin-derived peptides do not permeabilize bacterial membranes and kill microorganisms irrespective of their charge.
Dermcidin (DCD) is a recently described antimicrobial peptide, which is constitutively expressed in eccrine sweat glands and transported via sweat to the epidermal surface. By postsecretory proteolytic processing in sweat the dermcidin protein gives rise to several truncated DCD peptides which differ in length and net charge. In order to understand the mechanism of antimicrobial activity, we an...
متن کاملStaphylococcus aureus mutant screen reveals interaction of the human antimicrobial peptide dermcidin with membrane phospholipids.
Antimicrobial peptides (AMPs) form an important part of the innate host defense. In contrast to most AMPs, human dermcidin has an anionic net charge. To investigate whether bacteria have developed specific mechanisms of resistance to dermcidin, we screened for mutants of the leading human pathogen, Staphylococcus aureus, with altered resistance to dermcidin. To that end, we constructed a plasmi...

Background:Brucella spp. are facultative intracellular pathogens that can cause chronic infections in many tissues and organs. Objectives: To investigate serum dermcidin, salusin-alpha, salusin-beta and TNF-alpha levels and their correlation with each other in patients with acute brucellosis. Methods: From 50 patients hospitalized upon...

Dermcidin (DCD), an antimicrobial peptide with a broad spectrum of activity against bacteria such as Propionibacterum acnes, is expressed constitutively in sweat in the absence of stimulation due to injury or inflammation. The aim of this study was to determine the relationship between DCD expression and acne vulgaris associated with P. acnes. The antimicrobial activity of recombinant full-leng...
